Canadine Synthase from Thalzctrum Tuberosum Cell Cultures Catalyses the Formation of the Methylenedioxy Bridge in Berberine Synthesis
نویسنده
چکیده
An enzyme system catalysing the formation of the methylenedioxy bridge at ring A of (S)-canadine [=(S)tetrahydroberberine] from (S)-tetrahydrocolumbamine has been detected in microsomal preparations from different Ranunculaceae and Berberidaceae cell cultures. The cytochrome P-450 enzyme complex has been partly characterized from a protoberberine alkaloid producing Thalictrum tuberosum L. cell line. The enzyme complex consisting of a microsomal associated oxidase with a cytochrome P-450 reductase has a pH optimum at pH 8.5 and a temperature optimum of 40’. The apparent K, values are 33 PM for NADPH and 11.5 PM for tetrahydrocolumbamine.

Molecular cloning and characterization of CYP719, a methylenedioxy bridge-forming enzyme that belongs to a novel P450 family, from cultured Coptis japonica cells.
Two cytochrome P450 (P450) cDNAs involved in the biosynthesis of berberine, an antimicrobial benzylisoquinoline alkaloid, were isolated from cultured Coptis japonica cells and characterized. A sequence analysis showed that one C. japonica P450 (designated CYP719) belonged to a novel P450 family. Further, heterologous expression in yeast confirmed that it had the same activity as a methylenediox...
متن کاملOptimization of yeast-based production of medicinal protoberberine alkaloids
BACKGROUND Protoberberine alkaloids are bioactive molecules abundant in plant preparations for traditional medicines. Yeast engineered to express biosynthetic pathways for fermentative production of these compounds will further enable investigation of the medicinal properties of these molecules and development of alkaloid-based drugs with improved efficacy and safety. Here, we describe the opti...
متن کاملCytotoxic Activity of Alkaloids from Papaver rhoeas growing in Lebanon
This paper represents the phytochemical properties of Lebanese Papaver rhoeas, from which five protopine alkaloids are isolated, namely; stylopine (1), canadine (2), sinactine (3), berberine (4), and epiberberine (5). This is the first report for the isolation of epiberberine (5) from the genus Papaver and canadine (2) from Papaver rhoeas, suggesting a new chemotype of Papaver rhoeas growing in...
متن کاملCharacterization of a flavoprotein oxidase from opium poppy catalyzing the final steps in sanguinarine and papaverine biosynthesis.
Benzylisoquinoline alkaloids are a diverse class of plant specialized metabolites that includes the analgesic morphine, the antimicrobials sanguinarine and berberine, and the vasodilator papaverine. The two-electron oxidation of dihydrosanguinarine catalyzed by dihydrobenzophenanthridine oxidase (DBOX) is the final step in sanguinarine biosynthesis. The formation of the fully conjugated ring sy...
متن کاملPLANT REGENERATION FROM MESOPHYLL PROTOPLASTS OF POTATO (SOLANUM TUBEROSUM L.) CULTIVAR DELAWARE USING SILVER THIOSULFATE (STS)
The use of in vitro potato shoot cultures for protoplast isolation is desirable for consistent protoplast qualities. Ethylene build-up in the culture vessels causes problems for leaf growth (e.g. decreasing leaf surface, dry weight and chlorophyll) in shoot culture. Fifty to one hundred µM Silver thiosulfate (STS) produced a larger leaf area as a source of plant material for protoplast isolatio...
